Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
Characterising a peptide of this size relies on a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact molecule from related impurities, while electrospray mass spectrometry confirms molecular mass and detects truncation or oxidation products. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates modified residues. Because the molecule carries a lipid chain, assays must also distinguish the correctly conjugated product from incompletely acylated species.
Peptides in this class degrade mainly through hydrolysis, oxidation, and aggregation. The lipid modification improves plasma residence time but can also promote self-association in aqueous solution at higher concentrations. Oxidation of methionine and deamidation of asparagine residues are common chemical liabilities that accumulate during storage. Stability studies therefore track purity loss, aggregate formation, and changes in receptor-binding potency over time under defined temperature and humidity conditions.

The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
On Banlieue Triste, Hangman's Chair addressed the same thematic elements but less metaphorically, taking a more straightforward approach. Some of Banlieue Triste's topics were addressed for the first time by the group, ultimately reflecting "real parts" of their lives and closely associated with a "very personal" album artwork featuring hidden references. During the Banlieue Triste era, they began to delve deeper into introspection and focus on themes fuelled by personal issues. The lyrics directly address topics such as the near-disappearance of a band member following an overdose in 2016, recounted in the song "04/09/16"; the loss of family in "Negative Male Child"; and mental health and its associated difficulties in "Touch the Razor". The instrumental "Sidi Bel Abbes" pays tribute to Azzouni. The band's album artwork and music videos invariably referenced Azzouni. Kerrang! described Banlieue Triste as an album that displays "a darkness that comes not from the wilderness that so often makes up doom, but a more gritty, claustrophobic urban setting". The single "Lost Brothel", taken from the EP Bus de Nuit (2019), aligns with the band's dark thematic elements: "Prostitution, night, suburbia and unhappy loves". In his analysis, Desgroux considered that the band's themes were rooted on either side of the boulevard périphérique (ring road) of Paris. On one side, he described Paris, with its "mysteries", "legends", stories and "myths"; on the other, the suburb, with its violence, "crude reality", boredom and "ugly and sad aesthetic".

=== Toxicity === TTX is extremely toxic. The material safety data sheet for TTX lists the oral median lethal dose (LD50) for mice as 334 μg per kg. For comparison, the oral LD50 of potassium cyanide for mice is 8,500 μg per kg, demonstrating that even orally, TTX is more poisonous than cyanide. TTX is even more dangerous if administered intravenously; the amount needed to reach a lethal dose by injection is 8 μg per kg in mice. The toxin can enter the body of a victim by ingestion, injection, or inhalation, or through abraded skin. Poisoning occurring as a consequence of consumption of fish from the order Tetraodontiformes is extremely serious. The organs (e.g., liver) of the pufferfish can contain levels of tetrodotoxin sufficient to produce the described paralysis of the diaphragm and corresponding death due to respiratory failure. Toxicity varies between species and at different seasons and geographic localities, and the flesh of many pufferfish may not be dangerously toxic. The mechanism of toxicity is through the blockage of fast voltage-gated sodium channels, which are required for the normal transmission of signals between the body and brain. As a result, TTX causes loss of sensation, and paralysis of muscles including the diaphragm and intercostal muscles, stopping breathing.
=== 7 August === Sudanese prosecutors charged Hemedti and 16 other TASIS leaders with committing war crimes in El Fasher and South Kordofan. === Lanthipeptides === Lanthipeptides are one of the most well-studied families of RiPPs. The family is characterized by the presence of lanthionine (Lan) and 3-methyllanthionine (MeLan) residues in the final natural product. There are four major classes of lanthipeptides, delineated by the enzymes responsible for installation of Lan and MeLan. The dehydratase and cyclase can be two separate proteins or one multifunctional enzyme. Previously, lanthipeptides were known as "lantipeptides" before a consensus was reached in the field. Lantibiotics are lanthipeptides that have known antimicrobial activity. The founding member of the lanthipeptide family, nisin, is a lantibiotic that has been used to prevent the growth of food-born pathogens for over 40 years.
=== Optical sensors === Polymer optical fibers have generated increasing interest in recent years. Because of low cost, ease of handling, long wavelength transparency, great flexibility, and biocompatibility, polymer optical fibers show great potential for short-distance networking, optical sensing and power delivery. Electrospun nanofibers are particularly well-suitable for optical sensors because sensor sensitivity increases with increasing surface area per unit mass. Optical sensing works by detecting ions and molecules of interest via fluorescence quenching mechanism. Wang et al. successfully developed nanofibrous thin film optical sensors for metal ion (Fe3+ and Hg2+) and 2,4-dinitrotoluene (DNT) detection using the electrospinning technique. Quantum dots show useful optical and electrical properties, including high optical gain and photochemical stability. A variety of quantum dots have been successfully incorporated into polymer nanofibers. Meng et al. showed that quantum dot-doped polymer nanofiber sensor for humidity detection shows fast response, high sensitivity, and long-term stability while requiring low power consumption. Kelly et al. developed a sensor that warns first responders when the carbon filters in their respirators have become saturated with toxic fume particles. The respirators typically contain activated charcoal that traps airborne toxins. As the filters become saturated, chemicals begin to pass through and render the respirators useless.
==== Europe ==== In the European Union, Subutex and Suboxone, buprenorphine's high-dose sublingual tablet preparations, were approved for opioid use disorder treatment in September 2006. In the Netherlands, buprenorphine is a list II drug of the Opium Law, though special rules and guidelines apply to its prescription and dispensation. In France, buprenorphine prescription by general practitioners and dispensed by pharmacies has been permitted since the mid-1990s as a response to HIV and overdose risk. Deaths caused by heroin overdose were reduced by four-fifths between 1994 and 2002, and the incidence of AIDS among people who inject drugs in France fell from 25% in the mid-1990s to 6% in 2010.

It is a synthetic peptide triagonist that engages GIP, GLP-1, and glucagon receptors. Investigational compounds in this class are studied for metabolic conditions rather than for a single organ system.
No regulatory agency has approved it for any indication. It remains an investigational product studied within controlled clinical trial programs.
A dual agonist engages two receptor targets, typically GIP and GLP-1. A triagonist adds glucagon receptor activity, which is intended to influence energy expenditure alongside appetite and glucose handling.
Mass spectrometry provides the most direct confirmation of molecular mass. Reversed-phase chromatography adds a retention-time signature that supports identity when compared against a reference standard. No single method is sufficient on its own.
