This is a working overview of glucagon receptor, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-07-28. Anything still debated is marked as such rather than presented as settled.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
Retatrutide is a synthetic peptide developed as a single molecule that activates three distinct hormone receptors: GLP-1, GIP, and glucagon. The compound carries the internal designation LY3437943 and was engineered by modifying the backbone of glucose-dependent insulinotropic polypeptide. Its sequence incorporates non-natural amino acids and a fatty acid side chain that extends circulation time. The triple-agonist design aims to combine appetite suppression, improved insulin response, and increased energy expenditure in one agent. Published reports describe it as an investigational product rather than an approved medicine.

Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Retatrutide is an investigational synthetic peptide designed to activate three distinct receptor systems within a single molecule. Its pharmacological profile combines activity at the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor. This arrangement places it within a broader class of agents often described as multi-agonists, which contrast with compounds that engage one or two targets. Research interest centers on whether simultaneous signaling produces effects that single-receptor agonists cannot achieve alone. A single molecular entity also simplifies manufacturing and delivery logistics compared with combining separate agents.
Mechanistic proposals link each receptor to a different physiological role. Activation of the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors is associated with reduced appetite, slower gastric emptying, and glucose-dependent insulin release. Glucagon receptor signaling, by contrast, is associated with increased energy expenditure and altered lipid handling, though it can also raise blood glucose. The design intent is to balance these contributions so that weight reduction is enhanced without unacceptable glycemic trade-offs. How well that balance holds across individuals is not fully resolved.
=== Professional === Lisa attended her first IFBB professional competition at the 2014 Tampa Pro, where she placed 16th place. In 2015, Alina Popa became her coach and started training at the Armbrust PRO Gym. Her training paid off when she went on to win the 2015 Omaha Pro, the first time a professional British female bodybuilder won an overall professional competition since 1999.
Due to its broad availability, speed, and relatively low cost, it continues to be a common first-line tool in radiologic evaluation. Despite advances in CT, MRI, and other imaging techniques, there are many conditions in which traditional radiographs remain helpful in diagnosis. These include arthritis, pneumonia, bone tumors, fractures, congenital skeletal anomalies, and certain types of kidney stones. Mammography and DXA are two applications of low-energy projectional radiography, used for the evaluation of breast cancer and osteoporosis, respectively.
==== January 2024 ==== On 2 January, Israel conducted an airstrike in the Dahieh neighborhood of Beirut, resulting in the assassination of Saleh al-Arouri, the deputy chairman of the Hamas political bureau, and the death of six other members. Al-Arouri was also responsible for the expansion of Hamas's activities in the Israeli-occupied West Bank, including attacks on Israelis. The assassination occurred one day before Hezbollah commemorated the fourth anniversary of the assassination of senior Iranian military commander Qasem Soleimani. On 6 January, Hezbollah launched approximately 40 rockets into northern Israel, describing it as an "initial response" to al-Arouri's killing. According to Israel, the rockets targeted a Air traffic Control Base near Mount Meron, causing significant damage to it. On 8 January, Israel assassinated Wissam al-Tawil, the deputy commander of Hezbollah's Redwan Force, whom it accused of carrying out the attack on Meron airbase two days earlier. In response to al-Tawil's killing, Hezbollah launched a drone attack on Israel's Northern Command headquarters in Safed on the following day, situated approximately 20 kilometres (12 mi) from the border. Ali Hussein Barji, the commander of Hezbollah's aerial forces in southern Lebanon who was reportedly responsible for the attack, was killed by an airstrike, according to Israel, alongside three other Hezbollah members, in Khirbet Selm shortly before al-Tawil's funeral began. However, Hezbollah denied that Barji was killed.
=== Legacy waste === Due to historic activities typically related to the radium industry, uranium mining, and military programs, numerous sites contain or are contaminated with radioactivity. In the United States alone, the Department of Energy (DOE) states there are "millions of gallons of radioactive waste" as well as "thousands of tons of spent nuclear fuel and material" and also "huge quantities of contaminated soil and water." Despite copious quantities of waste, in 2007, the DOE stated a goal of cleaning all presently contaminated sites successfully by 2025. The Fernald, Ohio site for example had "31 million pounds of uranium product", "2.5 billion pounds of waste", "2.75 million cubic yards of contaminated soil and debris", and a "223 acre portion of the underlying Great Miami Aquifer had uranium levels above drinking standards." The United States has at least 108 sites designated as areas that are contaminated and unusable, sometimes many thousands of acres. The DOE wishes to clean or mitigate many or all by 2025, using the recently developed method of geomelting, however the task can be difficult and it acknowledges that some may never be completely remediated. In just one of these 108 larger designations, Oak Ridge National Laboratory (ORNL), there were for example at least "167 known contaminant release sites" in one of the three subdivisions of the 37,000-acre (150 km2) site. Some of the U.S. sites were smaller in nature, however, cleanup problems were simpler to address, and the DOE has successfully completed cleanup, or at least closure, of several sites.

Territories on both sides of the new Polish-Ukrainian border were also "ethnically cleansed". Of the Ukrainians and Lemkos living in Poland within the new borders (about 700,000), close to 95% were forcibly moved to the Soviet Ukraine, or (in 1947) to the new territories in northern and western Poland under Operation Vistula. In Volhynia, 98% of the Polish pre-war population was either killed or expelled; in Eastern Galicia, the Polish population was reduced by 92%. According to Timothy D. Snyder, about 70,000 Poles and about 20,000 Ukrainians were killed in the ethnic violence that occurred in the 1940s, both during and after the war. According to an estimate by historian Jan Grabowski, about 50,000 of the 250,000 Polish Jews who escaped the Nazis during the liquidation of ghettos survived without leaving Poland (the remainder perished). More were repatriated from the Soviet Union and elsewhere, and the February 1946 population census showed about 300,000 Jews within Poland's new borders.[e] Of the surviving Jews, many chose to emigrate or felt compelled to because of the anti-Jewish violence in Poland. Because of changing borders and the mass movements of people of various nationalities, the emerging communist Poland ended up with a mainly homogeneous, ethnically Polish population (97.6% according to the December 1950 census). The remaining members of ethnic minorities were not encouraged, by the authorities or by their neighbors, to emphasize their ethnic identities.[i][a1]
Veratridine is a steroidal alkaloid found in plants related to lilies, specifically the genera Veratrum and Schoenocaulon. Upon absorption through the skin or mucous membranes, it acts as a neurotoxin by binding to and preventing the inactivation of voltage-gated sodium ion channels in heart, nerve, and skeletal muscle cell membranes. Veratridine increases nerve excitability and intracellular Ca2+ concentrations.
== Function == The cell bodies produce the peptide hormone vasopressin, which is also known as anti-diuretic hormone (ADH), and the peptide hormone oxytocin. Both of these peptides are released from the posterior pituitary. ADH travels via the bloodstream to its target cells in the papillary ducts in the kidneys, enhancing water reabsorption. Oxytocin travels via the bloodstream to act at the mammary glands and the uterus. In the cell bodies, the hormones are packaged in large, membrane-bound vesicles that are transported down the axons to the nerve endings. The secretory granules are also stored in packets along the axon called Herring bodies. Similar magnocellular neurons are also found in the paraventricular nucleus.

== Adverse effects == There are limited amounts of research available on the effects of 3-CMC. The effects are likely comparable to those of other cathinones of which it is known exposure can result in symptoms such as tachycardia, hypertension, and episodes of psychosis. Users also report other side effects including an increase in body temperature, sweating, anxiety, and dry mouth.
=== Hair growth === Dermal macrophages are the essential component of the hair follicle immune system. They generally facilitate hair growth. There are three steps of hair growth: anagen, catagen, and telogen. During catagen, dermal macrophages' population decreases progressively when the hair grows. The number of dermal macrophages in hair follicles reaches a minimum during telogen (resting state) and increases during anagen (beginning of another growth cycle). In rodents, perifollicular macrophages can actively remove collagen fibres around the follicles via phagocytosis. This phenomenon might contribute to remodelling the follicular composition during anagen when dermal macrophages prevent the activation of follicular stem cells, thereby preventing entry to catagen, a process in which hair stops growing. Hence, dermal macrophages facilitate the growth of the hair by preventing the halt of the growth process and its regression. When under physical stress, the release of CCL2 (cytokine) in the hair follicle induces the infiltration of macrophages. The infiltrated macrophages mainly express an M1 phenotype, which are pro-inflammatory macrophages that could trigger apoptosis of cells in the follicle by their upregulation of pro-inflammatory cytokines such as TNF-a. However, TNF-a is a major factor facilitating hair regeneration by promoting various pathways' signalling (Wnt, NF-κB pathway). Dermal macrophages could also contribute to hair regeneration by skewing towards the anti-inflammatory phenotype (M2) under minor stress.
Enterovirus Hepatitis A is distinguished from other viral causes by its prolonged (2–6 week) incubation period and its ability to spread beyond the stomach and intestines into the liver. It often results in jaundice, or yellowing of the skin, but rarely leads to chronic liver dysfunction. The virus has been found to cause infection due to the consumption of fresh-cut produce which has fecal contamination. Hepatitis E Norovirus Rotavirus
In addition to altering its acidity, the presence of carbon dioxide in water also affects its electrical properties. When carbon dioxide dissolves in desalinated water, the electrical conductivity increases significantly from below 1 μS/cm to nearly 30 μS/cm. When heated, the water begins to gradually lose the conductivity induced by the presence of
==== Gene knockout studies ==== The large number of mouse compared to human FPR receptors makes it difficult to extrapolate human FPR1 functions based on genetic (e.g. gene knockout or forced overexpression) or other experimental manipulations of FPR receptors in mice. In any event, targeted disruption of the Fpr1 gene reduced the ability of mice to survive intravenous injection of the bacterial pathogen, listeria monocytogenes; disruption of the Fpr2 gene in mice produce a similar effect while disruption of both genes further lowered the survival of mice to the listeria challenge. The effect of these gene knockouts appeared due to faulty leukocyte function and other causes leading to a breakdown in the innate immune response. The functions of the human FPR1 receptor may be equivalent to the overlapping functions of the mouse Fpr1 and Fpr2 functions and therefore be critical in the defense against at least certain bacteria. Targeted disruption of FPR-rs1 produced a 33% reduction in the lifetime of mice; there was no specific pathology associated with this reduction.

Retatrutide is an investigational peptide that activates the GLP-1, GIP, and glucagon receptors at the same time. It is being studied for obesity and type 2 diabetes and has not been approved for clinical use. The internal code LY3437943 refers to the same molecule.
Most approved incretin therapies target one or two receptors, while retatrutide engages three. The addition of glucagon receptor activity is intended to raise energy expenditure. This broader mechanism is the subject of ongoing research rather than established practice.
No regulatory agency has approved retatrutide for any indication as of available public information. Development remains in later-stage trials. Availability claims found online should be treated as unverified.
